MOTS-c: the one my body voted against

Three starts, three stops. The last one ended after a second reaction, and that is the end of the experiment.

Goal: Insulin sensitivity and endurance, timed around training.

The decision

MOTS-c is the most exciting compound on paper in my whole stack and one of the least tested in people. Our own grade is a D on evidence. I knew that going in and ran it anyway, because the mechanism fit what I was after: better glucose handling and more in the tank for long training days.

It had already given me trouble once before this summer, so I put it last in line. Everything else had to be stable first. When I did restart in late July, I ran it fasted in the morning, three times a week, and built toward about 10 mg a week.

In October I came back to it at a lower dose: 2.5 mg twice a week, 5 mg total, on days that never overlap with my other shots so I could tell what was doing what.

What happened

I came off it in September with everything else except two compounds. Within days of restarting at the lower dose on October 8, I had another robust reaction in my leg, at the injection site, a big histamine-type response. Two reactions on one compound is a pattern, not bad luck.

On October 12 I took it off the list. Only a few doses had run, so nothing else in the stack was muddied by it.

Timeline

  1. Jun 2026

Parked at the back of the line. It had caused problems before.

  2. Jul 26, 2026

Restarted on fresh vials, fasted mornings, three times a week.

  3. Jul 30, 2026

Target set at about 10 mg a week (3.3 mg, Tue/Thu/Sat).

  4. Sep 21, 2026

Off, along with everything except retatrutide and GHK-Cu.

  5. Oct 8, 2026

Back on at a lower dose: 2.5 mg Mon and Thu.

  6. Oct 12, 2026

Off the list after a second robust histamine-type reaction at the injection site.
